The BCL6 gene, which functions as a transcription repressor, is the target of multiple chromosomal translocations in non-Hodgkin's lymphomas (NHL).The levels of expression of the BCL6 gene and protein have been demonstrated to predict the clinical outcome of diffuse large B-cell lymphomas.
Specimen Requirements
Collection:
The ordering pathologist will decide whether to order a conventional FISH preparation or a B-cell enrichment (BCE) FISH preparation.
Bone Marrow Aspirate: 1~2mL in Green top (Sodium Heparin) tube OR Lavender top (EDTA) tube. B-cell enrichment eligible.
Peripheral Blood: 2~5mL Green top (Sodium Heparin) tube OR Lavender top (EDTA) tube. B-cell enrichment eligible.
Fresh, Unfixed Tissue: Tissue in RPMI. A conventional FISH preparation will be performed as B-cell enrichment can not be performed on fresh tissues.
Fluids: Equal parts RPMI to specimen volume. A conventional FISH preparation will be performed as B-cell enrichment can not be performed on fresh fluid tissues.
Paraffin Block: H&E slide and paraffin block. B-cell enrichment can not be performed on paraffin embedded tissues.
Cut Slides: H&E slide and 1 unstained slide (cut at 4 microns) per probe. B-cell enrichment can not be performed on paraffin embedded tissues.
