Test Description and clinical significance

Genes
CSF3R

Disease:
chronic neutrophilic leukemia (CNL) and atypical chronic myeloid leukemia (aCML).

Colony Stimulating Factor 3 Receptor (CSF3R) encodes the receptor for granulocyte colony-stimulating factor (G-CSF), which regulates granulocyte proliferation and differentiation. Activating and truncating mutations in CSF3R are recurrent in myeloid malignancies, particularly chronic neutrophilic leukemia (CNL) and atypical chronic myeloid leukemia (aCML). Identification of CSF3R mutations can aid in diagnosis, provide prognostic information, and suggest therapeutic options, including JAK inhibitors and other targeted agents. This assay detects clinically relevant mutations in CSF3R, including activating variants in the extracellular domain and truncating variants in the cytoplasmic tail.

Specimen Requirements

Preparation:
PB, BM

Collection:
At least 0.5 ml EDTA, citrate (ACD) or Heparinized whole blood or bone marrow aspirates for DNA mutations

Stability:
96hours/4 to 7 days refrigerated

Unacceptable Conditions:
Specimen submitted is in the incorrect fixative or anticoagulant. Specimen received after the stability date. Gross hemolysis. Frozen specimen. Insufficient specimen. Unlabeled. Left un-refrigerated for an extended period.

Storage & Transport

Blood or bone marrow specimens should be kept at room temperature and preferably received within 96 hrs of drawing. Samples that are 4 to 7 days old should be refrigerated.
